Overview
Dermorphin is a naturally occurring opioid heptapeptide first isolated from the skin of South American Phyllomedusa frogs. Its defining feature is a D-alanine at position 2 — one of the earliest known examples of a ribosomally-derived vertebrate peptide bearing a D-amino acid — and that single stereochemical inversion is what gives dermorphin both its resistance to peptidase degradation and its very high affinity and selectivity for the μ-opioid receptor. In research it serves as a potent, selective μ-agonist tool for opioid-receptor pharmacology and peptide-transport questions. Natural ribosomal synthesis builds L-amino-acid peptides, so a D-residue is unusual and consequential. In dermorphin the D-Ala at position 2 blocks the aminopeptidase cleavage that would rapidly destroy an all-L sequence, and it locks the backbone into the conformation that binds the μ-opioid receptor with high affinity and selectivity. An accidentally all-L version of the same sequence is a different, far less active molecule — which is why stereochemical confirmation belongs on the COA.
Dermorphin is used as a selective μ-opioid-receptor agonist tool in receptor-pharmacology and analgesia research, and as a model substrate in studies of D-amino-acid peptide stability and blood-brain-barrier peptide transport.
