Mitochondrial-derived cytoprotective 24-mer peptide
Overview
Humanin was the first mitochondrial-derived peptide (MDP) to be described — encoded not in the nuclear genome but within the mitochondrial 16S rRNA (MT-RNR2) region. It was discovered through screens for factors that protect neurons against Alzheimer's-associated toxicity, and the research field around it centers on cytoprotection, apoptosis suppression, and metabolic signaling, positioning humanin alongside MOTS-c as a founding member of the mitochondrial-derived-peptide class. The commonly studied form is the 24-residue cytosolic sequence. PeptideXpo supplies humanin as a lyophilized powder at ≥ 98% HPLC. Researchers should note that several potency-enhanced analogs exist — most notably the S14G variant (HNG, 'Humanin-G'), reported to be dramatically more potent than wild-type in cytoprotection assays — so specifying wild-type versus HNG matters when comparing results across the literature. Mitochondrial-targeted peptides ship primarily to research labs studying OXPHOS, ROS biology, and mitochondrial dysfunction in disease models. Mitochondrial-derived peptides (MDPs) are small peptides encoded within the mitochondrial genome rather than nuclear DNA — a discovery that expanded the known coding capacity of mitochondria. Humanin was the first MDP identified (in the MT-RNR2 16S rRNA region) and established the concept; MOTS-c followed. HNG, or Humanin-G, carries a single serine-to-glycine substitution at position 14.
